چکیده

Cholera toxin and other bacterial toxins can induce electrogenic chloride (Cl) secretion in the small intestine resulting in secretory diarrhoea, when the colonic water reabsorption capacity is overwhelmed. The mechanism underlying this phenomena is that, these toxins increase intracellular cGMP and/or cAMP level through activation of guanylyl and adenylyl cyclase leading to the phosphorylation of the apical chloride channel (CFTR) and electrogenic Cl secretion as revealed in vitro by an increase in short-circuit current reflecting an increase in electrolyte transport in the intestine. The aim of the study was to investigate the effects of D-(+) glucose on water and electrolyte movements across rat jejunum after challenging with cholera toxin and dbcAMP (a lipophilic analog of cAMP which readily crosses the basolateral membrane of small intestinal cells); and also to investigate whether the magnitude of response to D-(+) glucose was related to the extent of secretion induced by dbcAMP. The measurement of the ion transport across the unstripped rat jejunum was carried out using Ussing chambers. The response to D-(+) glucose was studied in both CT-treated and untreated tissue; the results showed no significant difference between Isc responses to D-(+) glucose in unstimulated and CT-stimulated rat jejunum (∆Isc = 45.3 ± 12.9 μA/cm versus 38.9 ± 13.9 μA/cm, n = 8), whereas, in the studies where tissues treated with dbcAMP instead of CT, results showed a small but significant difference in D-(+) glucose response affected by dbcAMP (∆Isc = 12.9 ± 4.7 μA/cm versus 24.0 ± 4.3 μA/cm, n = 8).

The effect of theophylline on glucose and fluid transport across the rat jejunum.

The net absorption of fluid and electrolytes by the mammalian small intestine may be changed to a state of net secretion by the administration of agents that elevate the cyclic AMP content of the mucosal epithelium, e.g. cholera toxin, theophylline, dibutyrylcyclic AMP [for reviews see Banwell &Sherr(1973),Sladen(1973), Field(1974) and Kimberg (1974)l. In contrast with the wealth of data on the...

5-Hydroxytryptamine and cholera secretion: a histochemical and physiological study in cats.

The effect of cholera toxin on the content of 5-hydroxytryptamine (5-HT) in the enterochromaffin cells of the cat small intestine was estimated by cytofluorimetry of individual enterochromaffin cells at varying times after exposing the intestinal mucosa to the toxin. The observed changes in 5-HT levels in the enterochromaffin cells were correlated with the simultaneously measured rate of net fl...

intestinal fluid and electrolyte secretion induced by cholera and Escherichia coli

Cholera toxin and Escherichia coli heat labile toxin (LT) induced intestinal secretion has in the past been attributed exclusively to an increase in intracellular cAMP whereas E coli heat stable toxin (ST) induced secretion is mediated through cGMP. Evidence is accumulating on the importance of 5-hydroxytryptamine (5-HT) in cholera toxin induced secretion, but its role in LT and ST is not well ...

Significance of abnormal rabbit ileal histology in the pathogenesis of diarrhea.

In spite of several macroscopic criteria for predicting the presence of histological abnormalities in rabbit ileum, microscopic ileal abnormalities still can escape detection. The effect of histologically abnormal rabbit ileum was evaluated on basal intestinal absorption, on basal absorption, on basal adenylate cyclase activity, and on cholera toxin-induced secretion and cholera toxin-induced s...

5-hydroxytryptamine release into human jejunum by cholera toxin.

BACKGROUND Cholera toxin produces intestinal secretion by activation of the adenylate cyclase complex. However animal studies have shown 5-hydroxytryptamine may be released after exposure to cholera toxin, and thereby contribute to the secretory state. AIM To determine whether cholera toxin releases 5-hydroxytryptamine in human jejunum. SUBJECTS Seven male subjects were given a subclinical ...
